Research progress on the drug resistance mechanisms of temozolomide in glioblastoma and long non-coding RNA-based countermeasures

Abstract:Glioblastoma(GBM)is a highly malignant tumor with a high recurrence rate,leading to poor prognosis in affected patients.As the first-line chemotherapeutic agent for GBM,temozolomide(TMZ)induces tumor cell apoptosis by causing DNA damage.However,drug resistance to TMZ has emerged as a critical factor affecting chemotherapy efficacy.Investigating and analyzing TMZ resistance mechanisms,with the aim of reversing resistance,represents a crucial strategy for enhancing chemotherapy outcomes.O6-methylguanine-DNA methyltransferase(MGMT),cancer stem cells,DNA damage repair systems,autophagy,the tumor immune microenvironment,and abnormalities in related gene expression and signaling pathways are all closely associated with TMZ resistance.With advancing clinical research,the role of long non-coding RNA(lncRNA)in tumor cell drug resistance has also garnered significant attention.This review summarizes the mechanisms of TMZ resistance in GBM and the progress in lncRNA research related to GBM and TMZ resistance,aiming to provide countermeasures based on lncRNA for overcoming TMZ resistance in GBM and to identify novel targets and strategies.
