Abstract:High-grade glioma (HGG) is one of the most aggressive primary brain tumors, especially diffuse midline glioma (DMG) with H3K27 mutations, which often has a median survival of less than 18 months [1]. Although there have been some advances in molecular subtyping and targeted therapy for HGG in recent years, effective treatment strategies for HGG with PDGFRA mutations are still lacking [2-3]. PDGFRA is one of the key driver genes in HGG, and its mutations or amplifications can lead to abnormal proliferation and survival of tumor cells, making the development of drugs targeting PDGFRA of significant clinical importance [3-5].
