Breaking Through the Brainstem禁区: Advances in CAR-T Cell Therapy for H3K27M-Mutant Diffuse Midline Glioma
Abstract:H3K27M-mutant (H3K27M+) diffuse midline glioma (diffuse midline glioma, H3K27M+ DMG) is a common malignant tumor of the central nervous system, predominantly occurring in children and adolescents [1]. Due to the tumor's location in the pons or deep brainstem, surgical intervention is not feasible. Even with aggressive treatments such as radiotherapy and chemotherapy, more than 90% of patients die within 18 months of diagnosis [2]. Recent preclinical studies have shown that GD2-targeted chimeric antigen receptor T cells (GD2-CAR-T) exhibit significant efficacy in treating DMG [3-5].
