Effects of silencing eukaryotic translation initiation factor 3E on T98G human glioblastoma cells

Abstract:Objective To investigate the effects of silencing eukaryotic translation initiation factor 3E(EIF3E)on T98G human glioblastoma cells and explore new and effective therapeutic approaches.Methods The EIF3E gene in T98G cells was silenced through siRNA or lentiviral transfection.The cells were divided into a si-NC control group and a si-EIF3E experimental group(with silenced EIF3E gene).Changes in cell cycle,invasion,migration,proliferation,and apoptosis were analyzed using flow cytometry and absorbance assays.Results Compared with the si-NC control group,the si-EIF3E experimental group exhibited inhibited proliferation and invasion of T98G cells(both P<0.05).The si-EIF3E experimental group arrested the cell cycle in the G0/G1 phase.Cell proliferation was inhibited in the si-EIF3E experimental group at 24,48 and 72 h post-transfection(all P<0.05).There was no statistically significant difference in the apoptosis rate between the si-EIF3E experimental group and the si-NC control group at 24h post-transfection(P>0.05).Conclusions The EIF3E gene may possess novel functions in controlling glioblastoma multiforme(GBM).Silencing the EIF3E gene can impede the proliferation,cell cycle progression,invasion,and metastasis of T98G cells,providing evidence for the tumor-suppressive role of silencing the EIF3E gene in GBM.However,there is currently no evidence to suggest that silencing the EIF3E gene promotes apoptosis in T98G cells.
