Study on metabolic characteristics of cerebrospinal fluid in patients with occult tethered cord syndrome based on liquid chromatography-tandem mass spectrometry

Abstract:Objective To analyze the metabolic characteristics,metabolic pathways,and action targets of cerebrospinal fluid(CSF)in occult tethered cord syndrome(OTCS).Methods The participants were divided into OTCS group(OTCS patients,n=15)and normal control group(n=15).The changes of CSF metabolites in the two groups were detected by liquid chromatography-tandem mass spectrometry(LC-MS/MS)technology,and the differentially expressed metabolites were analyzed by bioinformatics.And the receiver operating characteristic(ROC)curve was used to verify the potential markers.Results The metabolomics results revealed that compared with the normal control group,there were 87 differentially expressed metabolites in the OTCS group,with 46 being upregulated and 41 downregulated.The differentially expressed metabolites in the OTCS patients mainly affect the occurrence of the disease through the pathways such as the TCA cycle,GABA synapses,glutamatergic synapses,and aminoacyl-tRNA biosynthesis.Among them,the upregulated succinic acid,L-glutamic acid and L-isoleucine,and the downregulated citric acid can be used as potential diagnostic markers for OTCS,demonstrating good diagnostic performance as verified by the ROC curve.Conclusions Metabonomics results indicate that there is abnormal cerebrospinal fluid metabolism in OTCS,and succinic acid,L-glutamic acid,L-isoleucine and citric acid are preliminarily used as potential markers for OTCS diagnosis.
