Role of HMGB1 in early edema after human cerebral contusion/laceration

Abstract:Objective To explore the role of high-mobility group box 1 (HMGB1) in early edema after human cerebral contusion/laceration (CCL).Methods Twenty patients with CCL were collected as experimental group and divided into 6 h (n =7),12 h (n =5),1 d (n =4),3 d (n =4) groups after CCL.The normal brain tissues from 5 patients were collected as control group (n =5).The expressions of HMGB1 and aquaporin 4 (AQP4) of the experimental and control groups were detected by immunofluorescence staining.Results Compared with control group,the ratio of HMGB1 transferred to the outside of the nucleus increased significantly 6 h after CCL (P <0.05) and reached peak 1 d after CCL(P < 0.05).Compared with control group,the expression of AQP4 increased significantly 6 h after CCL (P < 0.05),continued to increase and reached peak 3 d after CCL(P < 0.05).The expression of AQP4 was found mainly located in astrocytes.Conclusion HMGB1 participates in the pathophysiologic process of brain edema after CCL.
