Effect of Ginkgo biloba extract on expressions of MMP 2 and MMP 9 after traumatic brain injury in rats

Abstract:Objective To investigate the effect of Ginkgo biloba extract (GBE) on expression of matrix metalloproteinase-2 (MMP-2) and MMP-9 after traumatic brain injury (TBI) in rats. Methods TBI animal models were established by Feeney method. Eighty SD rats were randomly and equally divided into 4 groups:sham operation group (Sham group), TBI group, TBI+GBE group and TBI+doxycycline (DOX) group. The modified neurologic severity score (mNSS) was tested on days 1, 3, 7 and 14 after modeling. Brain water content was measured by wet-dry weight method. The MMP-2 and MMP-9 expressions were determined by immunohisto-chemical staining and Western blotting. Results Compared with the Sham group, the levels of brain water content and mNSS significantly increased (P<0.05), and the protein expressions of MMP-2 and MMP-9 were up-regulated in others 3 group (P<0.05). Compared with the TBl group, brain water content and mNSS significantly decreased (P< 0.05), and the protein expressions of MMP-2 and MMP-9 were down-regulated in TBI+GBE group and TBI+DOX group (P<0.05). Conclusions GBE can reduce brain edema and improve neurological function after TBI, and its mechanism may be related to inhibiting MMP-2 and MMP-9 expressions.
