Research of relationship between Wip1 gene and DNA nucleotide excision repair in glioma cells

Abstract:Objective To investigate the influence of Wip1 gene silencing on DNA nucleotide excision repair enzymes XPA and XPC in U251 glioma cells.Methods Glioma U251 cells were cultured in vitro and infected with Wip1 shRNA lentiviral vector as Wip1 (-) group,which was confirmed by qRT-PCR.The cells were infected by negative control lentiviral vector as NC group.Then the cells were treated with TMZ and divided into NC group,NC+TMZ group,Wip1 (-) group and Wip1 (-)+TMZ group.The cell proliferation was detected by CCK-8 kit.The expressions of XPA and XPC mRNA were detected by qRT-PCR.The expressions of XPA and XPC proteins were detected by Western blot.Results The proliferation rate was lower in Wip1(-)+TMZ group than in the other groups 48,72,96,120,and 144 h after TMZ treatment (P < 0.05).The expressions of XPA and XPC mRNA were higher in NC+TMZ group than in NC group 48 h after TMZ treatment (P < 0.05).The expressions of XPA and XPC proteins were higher in NC+TMZ group than in NC group 48 h after TMZ treatment (P< 0.05).The expressions ofXPC mRNA and XPA protein were higher in Wipl(-)+TMZ group than in Wip1 (-) group (P < 0.05).Conclusion Wip1 gene is related to DNA injury and repair in U251 glioma cells,can regulate downstream mediators XPA and XPC to participate in the process of cellular nucleotide excision repair.
