Effect and mechanism of dihydroartemisinin on migration and invasion of glioma U87 cells

Abstract:Objective To investigate the effects of dihydroartemisinin (DHA) on migration and invasion in U87 glioma cells. Methods The U87 cells were cultured with different concentrations of DHA, and CCK8 assay was performed to evaluate the cell survival. Cell migration and invasive capacities were evaluated by means of scratch wound assay and transwell matrix penetration assay. The activity and expressions of invasion-related proteins matrix-metallopeptidase 2 ( MMP-2 ) and MMP-9 were analyzed by Western blotting . Results DHA caused concentration-dependent and time-dependent inhibition of the proliferation of U87 cells (P<0.05). Cells treated with DHA showed significantly decreased U87 cells migration and invasion capabilities. Western blotting demonstrated that the protein expressions of MMP-2 and MMP-9 were suppressed by DHA in U87 cells (P<0.05). Conclusions DHA may play its role inhibiting glioma cell line U87 migration and invasion via suppressing the expressions of MMP-2 and MMP-9.
