A study on the protective effect of GDF-15 during heart transplantation

Abstract:Objective To investigate the protective effect of growth differentiation factor-15(GDF-15)in heart transplantation.Methods C57BL/6 mice of the same genotype and transgenic mice with high expression of GDF-15 were used,and their hearts were transplanted into the abdominal cavity of the same genotype mice 24 hours later.Immunohistochemistry,gene expression,and fibrosis detection were performed on the hearts of the recipient mice.Results After heart transplantation in mice,the apoptosis of myocardial cells in the high expression group of GDF-15 was significantly decreased compared to the control group(P<0.05).The expression of various pro angiogenic factors in the high expression group of GDF-15 was significantly increased compared to the control group(P<0.05).The inflammatory factors IL-6 and IL-1β,TNF-γ in the transplanted tissues of the high expression group of GDF-15 were also found to be elevated.The expression of GDF-15 was significantly reduced compared to the control group(P<0.05),and the number of T regulatory cells(Treg CD4,CD25,FoxP3 positive)in the high expression group of GDF-15 was significantly increased compared to the control group(P<0.05).Conclusion High expression of GDF-15 can provide a certain protective effect on transplanted hearts.
