2,3,5,4′-Tetrahydroxystilbene-2-O-β-D-Glucoside protects hearts from iso-proterenol-induced cardiac hypertrophy in mice

Abstract:[ Abstract]:Objective To explore the anti-cardiac hypertrophy ( induced by isoproterenol) effect of 2,3,5,4′-Tetrahydroxys?tilbene-2-O-β-D-Glucoside( TSG) in C57 BL/6 mice. Methods Forty C57 BL/6 mice were randomly divided into four groups ( n=10 in each group):Sham group;TSG;ISO;ISO+TSG. We created the mouse cardiac hypertrophy model through subcutaneous injection of isoproterenol (5 mg/kg per day, for 1 week). Echocardiography was used to determine cardiac function. HW/BW and PW/BW were used to measure the degree of cardiac hypertrophy. HE staining was used to measure the mean cross-sectional area of cardiomyocytes in each group. RT-PCR and Western blotting were used to determine the mRNA and protein levels of ANP,α-MHC and β-MHC. Re?sults TSG treatment significantly improved the cardiac functional recovery and reduced HW/BW, LW/BW and the mean cross-sec?tional area of cardiomyocytes. Additionally, TSG treatment significantly reduced the expression of the mRNA and protein levels of ANP, andβ-MHC, while improved the expression of the mRNA and protein levels ofα-MHC. Conclusion Our results shows that TSG pos?sesses cardioprotective effects against isoproterenol-induced cardiac hypertrophy in mice.
