Abstract:Alzheimer's disease(AD)is a progressive neurodegenerative disorder characterized by β-amyloid deposition, hyperphosphorylated tau pathology, and neuronal loss, which could begin 10-20 years before onset of clinical symptoms.Advances in biomarker research and the concept of the cognitive continuum have shifted the diagnosis of AD from a clinically based framework to a biology-centered approach.Blood-based and neuroimaging biomarkers now enable early screening and diagnosis, and predict disease progression and prognosis.Furthermore, the trajectory of AD cognitive continuum is influenced by genetic factors, comorbidities, lifestyle, and cognitive reserve.Multimodal interventions,anti-amyloid immunotherapies, digital tools, and neuromodulation devices offer promising strategies for early interventions, contributing to delayed disease progression and improved outcomes.
