Association between insomnia and type 2 diabetes:a bidirectional two-sample Mendelian randomization study

Abstract:Objective To explore the association between insommia and type 2 diabetes(T2DM)by using Mendelian randomization(MR).Methods Summary data from large-scale population-based genome-wide association studies(GWAS)was used.The inverse variance weighted(IVW)method was used as the primary analysis approach to evaluate the causal effects.Sensitivity analyses,including weighted median(WM)method,MR-Egger regression method and leave-one-out method et al,were used to verify the reliability of the MR analysis results.Results The analysis of IVW demonstrated that insomnia could increase the risk of T2DM(IVW:OR 1.10,95%CI 1.06-1.14,P<0.001).The results were supported by the WM and MR-Egger regression method(WM:OR 1.09,95%CI 1.06-1.13,P<0.001;MR-Egger:OR1.19,95%CI 1.03-1.37,P<0.05).Reverse MR analysis showed no relationship between T2DM and increased risk of insomnia(IVW:OR 1.01,95%CI 1.00-1.03,P=0.058).There was no horizontal pleiotropy in instrumental variables(IV).Sensitivity analysis suggested that MR analysis results were stable.Conclusion The results demonstrate that insomnia is a risk factor for T2DM.while T2DM has no significant effect on the risk of insomnia.
