The relationship between drug resistance and the methylation status of wnt antagonist family genes in ovarian cancer
Abstract:Objective To investigate the relationship between drug resistance and the methylation status of Wnt antagonist family genes in ovarian cancer, and explore high effective way to reverse drug resistance. Methods The expression of the promoter methylation status and mRNA expression of Wnt antagonist family genes in SKOV3, OVCAR-8, A2780 and A2780/D cells were detected by methylation-specific PCR (MSP) and RT-PCR in West China Second Hospital in 2008. MTT was used to check the reversion of drug resistance in OVCAR-8 cell after treated with the demethylating agent, 5-aza-2'-deoxycytidine and then, the promoter methylation status and mRNA expression of WIF-1 were detected again. Results The methylation level of WIF-1 was significantly higher in OVCAR-8 cell than in other cells and might correlated with taxol resistant. 5-aza-2'-deoxycytidine could reactivate the expression of WIF-1 and reverse drug resistance. Conclusion The methylation status of WIF-1 plays an important role in the response to taxol in OVCAR-8 cell, and demethylations maybe reverse the drug resistant partially.
