Effect of IL-6 on androgen activity and its role in the polycystic ovary syndrome pathogenesis

Abstract:;Objective To study the effect of IL-6 on the secretion of testosterone in theca interna cells (TI) and on the expression of androgen receptor ( AR) in ovarian granular cells( CC) and to explore the role of interleukin 6 (IL-6) in the polycystic ovary syndrome pathogenesis. Methods Different concentrations of IL-6( 10ng/L、100ng/L、1000ng/L) were administered into cells medium of TI and GC. Secretion of testosterone in TI and expression of androgen receptor (AR) mRNA in GC were detected. Results The testosterone levels have no difference between the study groups (10ng/L,100ng/L,1000ng/L)and the control group(0ng/L)at 24h,48h,72h in the TI. The mRNA expressions of AR in GC were upregulated in groups with 100ng/L and 1000ng/L IL-6 at 72h by RT-PCR. Conclusion Although IL-6 can t induce testosterone secretion directly, but it can upregulate the mRNA expression of AR, so IL-6 can improve androgen activity indirectly, which probably can explain the phenomenon; chronic subclinical inflammation may result in PCOS.
