The expression of micro RNA in placenta from severe preeclampsia patients

Abstract:Objective To investigate differential expression of microRNA in the human placentae between severe preeclampsia and normal pregnancy and the relationship between microRNA and pathogenesis of severe preeclampsia.Methods Placental tissues were obtained from twenty-four pregnancies complicated by severe PE and twenty-six normal pregnant women at the time of caesarean section who were hospitalized in Department of Gynecology and Obstetrics of Nanjing Drum Tower Hospital,the Affiliated Hospital of Nanjing University Medical School between March,2005 and August,2008.Using microarray screen the differential expression of microRNA in the severe preeclampsia(4 specimens)and normal pregnancy placentae(4 specimens),and then some markable differential microRNAs were validated with quantitative real-time polymerase chain reaction in twenty-four severe preeclampsia pregnant,and twenty-six normal pregnant women placentae.Results Compared with normal placenta,we found that expression of miR-130a,miR-181a,miR-222,miR-16,miR-26b,miR-29b and miR-195 in severe preeclampsia placentae was significantly high compared with those in control,while expression of miR-612,miR-491-5p,miR-335,miR-519d,miR-520g,miR-126 and miR-451 was no significant.Conclusion The differential micmRNA(miR-130a,miR-181a,miR-222,miR-16,miR-26b,miR-29b and miR-195)may involve in pathogenesis of severe preeclampsia.
