Expression and significance of the gap junction protein connexin 43 in congenital hydronephrosis ureteropelvic junction stenosis segment

Abstract:Objective To study the expression of gap junction protein connexin 43 and Ki-67 in the stenotic tissue of congenital ureteropelvic junction obstruction in children and the relationship between them and the pathogenesis of con-genital hydronephrosis. Methods Case groups biopsies from forty pyeloplasty specimens were included in the study, which were sectioned through Anderson- Hynes and stained for connexin 43. These patients were confirmed by imaging and B-mode ultrasonic examination. Ten biopsies with normal ureteropelvic junction including 6 cases of Wilms tumor tissue , 3 cases of renal hypoplasia and one case of normal UPJ tissue specimens were selected as control group. Immu-nohistochemistry methods were used to detect the expression of Cx43 and Ki-67 in ureteropelvic junction wall. Results Gap junction Cx43 and Ki-67 were expressed in case and control groups.They were localized to between the circular muscle and longitudinal muscle. Expression of Cx43 in case group was lower than in control group (P < 0.05). Expres-sion of Ki-67 in case group was significantly higher than control group(P < 0.05), the difference being of statistical sig-nificance (P < 0.05 ).Conclusion The lack of connexin 43 may lead to the abnormal spontaneous dynamic conduction and cell proliferation in ureteropelvic junction stenosis segment.
