Expression and clinical significance of BCL2A1 in glioma

Abstract:Objective To investigate the expression and clinical significance of BCL2A1 in glioma.Methods Transcriptomic data and clinical information of glioma patients were obtained from the TCGA database.The R package"limma"was used to analyze BCL2A1 expression differences between tumor and adjacent tissues.Kaplan-Meier survival analysis was performed to evaluate the correlation between BCL2A1 expression(at both transcriptional and protein levels)and patient prognosis.Univariate and multivariate Cox regression analyses were applied to assess the association of BCL2A1 expression with survival outcomes.In vitro experiments using glioma cell lines U251 and U87 MG were conducted:BCL2A1 expression was knocked down via specific siRNA transfection,followed by CCK-8 assay for proliferation analysis,flow cytometry for cell cycle detection,and Transwell assay for migration evaluation.Results Database analysis revealed significantly higher BCL2A1 transcriptional levels in glioma tissues compared to adjacent normal tissues(P<0.01).High BCL2A1 expression was identified as an independent risk factor for poor prognosis in glioma patients.Functional experiments demonstrated that BCL2A1 knockdown significantly inhibited cell proliferation capacity(P<0.001),induced G0/G1 phase cell cycle arrest,and markedly reduced cell migration ability.Conclusion Our findings suggest that BCL2A1 serves as a novel prognostic biomarker in glioma,and its overexpression drives tumor progression by promoting cell proliferation and migration.
