Abstract:Objective To investigate the effects of genoside Rh2 on the apoptosis of human brain glioma cells and its mechanism. Methods Human brain glioma cells U87MG were randomly divided into two groups, i.e. experimental group in which U87MG cells were cultured in the medium containing 20 μg/ml genoside Rh2 and control group in which U87MG cells were cultured in the medium containing isodose physiological salt solution. The apoptosis of U87MG cells was determined by flow cytometry. And then the intracellular calcium concentration of U387MG cells was measured by laser scanning confocal microscope and flow cytometry. Results The rate of U87MG cells apoptosis and intracellular calcium concentration were significantly higher in the experimental group than those in the control group (P<0.05). Moreover the intracellular calcium concentration and the apoptosis rate of U87MG cells raised by genoside Rh2 were significantly decreased by nifedipine [(a specitic inhibitor of Ca++channel (P<0.05)]. Conclusion Genoside Rh2 can promote apoptosis of human brain glioma U87MG cells by the up-regulation of intracellular calcium concentration.
