Abstract:Objective To study the effect of curcumin on invasion and migration of human glioma cell line SHG44 and its mechanism. Methods CCK-8 kit assay was used to assess the effects of curcumin (0, 5, 10, 20, 40μmol/L) on the proliferation of human glioma cell line SHG44 24, 48, 72 hours after the action of curcumin. The cell scratch test and Transwell assay were used to evaluate the effects of curcumin on the invasion and migration of SHG44 cells. Western blotting was used to detect the expressions of matrix metalloproteinase (MMP-2) and MMP-9 in SHG 44 cells. Results CCK-8 kit assay showed that the curcumin significantly inhibited the proliferation of SHG44 with a concentration and time-dependent manner. The invasion and migration of SHG44 cells exposed to curcumin were significantly suppressed compared with the control group (P<0.05). The curcumin significantly reduced the expression levels of MMP-2 and MMP-9 in SHG 44 cells compared with the control group (P<0.05). Conclusions Curcumin can inhibit the proliferation, invasion and migration of human glioma cell line SHG44 with a concentration and time-dependent manner. The mechanism may involve in the down regulation of MMP-2 and MMP-9 in SHG 44 cells.
