Expressions of miR-9 and PPARγ in human brain gliomas and their clinical meanings

Abstract:Objective To investigate the expressions of miR-9 and peroxisome proliferator activated receptor γ (PPAR γ) in human brain gliomas and their clinical meanings.Methods The levels of miR-9 and PPARγ mRNA expressions were determined by qRT-PCR and the level of PPAR γ protein expression was determined by immunohistochemical technique in 76 specimens of brain glioma tissues derived from the patients who were surgically treated in our hospital from February, 2013 to February, 2015, and 39 specimens of normal brain tissues derived from the patients with brain injury who were surgically treated in our hospital during the same period. The factors related to the prognoses were analyzed by COX regression model analysis. Results The level of miR-9 mRNA expression was significantly higher in the glioma tissues than that in the normal brain tissues (P<0.05). The level of PPAR γ mRNA expression and the positive rate of PPARγ protein expression were significantly lower in the glioma tissues than those in the normal brain tissues (P<0.05). The positive rate of miR-9 mRNA expression was significantly higher and the positive rate of PPAR γ mRNA expression was significantly lower in WHO gradesⅢ~Ⅳglioma tissues than those in WHO gradesⅠ~Ⅱglioma tissues(P<0.05).The independent risk factors related to the prognoses in the patients with gliomas included over expression of miR-9,low expression of PPAR γ,high WHO grade glioma and incomplete resection of the tumors.The expression of miR-9 was significantly related negatively to that of PPAR γ expression in the glioma tissues (r=-0.573, P<0.05). Conclusion WHO grade of the tumor and prognoses are closely to expressions of miR-9 and PPARγ in the tumorous tissues in the patients with gliomas.
