Effects of miRNA-145 on proliferation and apoptosis of U251 glioma cells

Abstract:Objective To investigate the effects of miRNA-145 on the proliferation and apoptosis of U251 glioma ceils.Methods Quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) was applied to detect the expression of miRNA-145 in 10 human peritumoral tissues and 10 glioma tissues,and 3 glioma cell lines and normal glial cells.The miRNA-145 mimics were transfected into U251 glioma cells.The proliferation and apoptosis of U251 glioma ceils was determined by Mosmann Tetrazolium test (MTT) assay and flow cytometry (FCM),respectively.Results The level of miRNA-145 expression in the glioma tissues was significantly lower than that in the peritumoral tissues (P<0.05).The expression levels of miRNA-145 in glioma cells of U251,U87 and T98 were significantly lower than normal glial cells line HEB (P<0.01).The transfection of miRNA-145 mimics into U251 glioma cells significantly increased the expression of miRNA-145 in U251 glioma cells,decreased U251 cell proliferation and induced U251 cell apoptosis (P<0.05).Conclusions The expression of miRNA-145 is significantly decreased in glioma tissues and U251 cells.The up-regulation of miRNA-145 can inhibit the proliferation and induce apoptosis of U251 cells.The miRNA-145 might be a new molecular target for the treatment of glioma.
