Abstract:High mobility group box 1(HMGB1)is a conserved non-histone nuclear protein that not only participates in intracellular processes such as DNA replication, recombination and repair, but also involves in inflammatory response extracellularly as a damage-associated molecular pattern(DAMP).Acute lung injury(ALI)is an acute diffuse pulmonary inflammatory response caused by various factors such as infection and trauma. Its main characteristics include the disruption of the alveolar-capillary barrier,non-cardiogenic pulmonary edema, and hypoxemia. With the continuous deepening of research in recent years,it has been discovered that HMGB1 plays an important role in ALI and regulates its occurrence and development. Therefore, this article summarizes the research on the mechanism of HMGB1 in ALI,and reviews how HMGB1 alleviates inflammatory responses, improves pulmonary edema and tissue damage by regulating autophagy, immune cell functions, and ferroptosis during the process of ALI.The aim is to explore the mechanism of HMGB1 intervention in ALI,in order to provide new ideas for protecting lung function and improving lung injury.
