Mechanism of action of cuproptosis on cardiomyocyte injury and related cardiovascular disease

Abstract:Copper is an essential mineral.Adequate amounts of copper are required to sustain various physiological processes,including the cardiovascular system.Copper-induced cell death is shown to be a novel non-apoptotic cell death mode.Excess copper promotes protein toxicity stress response through copper-dependent abnormal oligomerization of lipoylated proteins in the tricarboxylic acid(TCA)cycle and reduces iron-sulfur cluster protein levels,which induces cuproptosis.Cuproptosis is increasingly recognized as an important process in mediating the pathogenesis and progression of cardiovascular diseases,including arrhythmias,heart failure,cardiomyopathy,and aortic aneurysm.Therefore,an insight into the mechanisms of cuproptosis in cardiovascular disease may help to improve the management of cardiovascular disease,as well as provide therapeutic ideas for cardiovascular diseases with copper-induced cell death.This article summarizes copper homeostasis-related mechanisms,the mechanisms of Cu2+ induced myocardial cell injury,cuproptosis induced cardiovascular disease,and the treatment of cuproptosis induced myocardial cell injury.
