The relationship of long chain non-coding RNA urothelial carcinoma-associated 1 gene with inflammatory factors,severity and prognosis in the septic patients

Abstract:Objective To explore the relationship of long chain non-coding RNA urothelial carcinoma-associated 1(lncRNA UCA1)gene with inflammatory factors,severity and 28-day mortality in the septic patients.Methods Clinical data of 174 patients with sepsis admitted to the 960th Hospital of the Joint Logistics Support Force of the People′s Liberation Army of China from January 2022 to December 2023 were collected.The healthy control group was matched 1 ∶ 1 with sepsis patients according to the same gender and age difference of 1 year with physical examinations during the same period.The level of lncRNA UCA1 in peripheral blood monocytes were detected by reverse transcription quantitative real-time polymerase chain reaction(RT-qPCR).Enzyme-linked immunosorbent assay(ELISA)method was used to detect the level of tumor necrosis factor(TNF)-α,interleukin-6(IL-6),IL-17,intercellular adhesion molecule 1(ICAM1),and vascular cell adhesion molecule 1(VCAM1)in serum.The COX risk proportional regression model was used to analyze the risk factors related to 28-day mortality in sepsis patients.Receiver operating characteristic(ROC)curves were used to evaluate the efficacy of independent risk factors in predicting 28-day mortality in sepsis patients.Results Compared with the control group,the patients with sepsis had higher levels of lncRNA UCA1(Z =76.235,P<0.001).In patients with sepsis,the level of lncRNA UCA1 were positively correlated with TNF-α(r =0.384),IL-6(r =0.308),IL-17(r =0.472),ICAM(r = 0.361),VCAM1(r =0.492),APACHEⅡ score(r =0.393),and SOFA score(r =0.427)(P<0.001).In the control group,there was no correlation between lncRNA UCA1 and the above parameters(P>0.05).Elevated levels of lncRNA UCA1(HR = 2.186,P = 0.007),increased age(HR = 1.245,P =0.011),fungal infection(HR =19.259,P =0.010),increased C-reactive protein(CRP)(HR =1.334,P =0.036)and APACHEⅡ score(HR =1.245,P =0.017)were independent risk factors for 28-day mortality in sepsis patients.The result of ROC curve analysis showed that lncRNA UCA1(AUC=0.757),APACHEⅡ score(AUC =0.865),CRP(AUC =0.731),and age(AUC =0.616)could predict the 28-day mortality in sepsis patients.Conclusions The elevated levels of lncRNA UCA1 in sepsis patients are associated with various inflammatory cytokines,disease severity,and poor prognosis.
