Research progress of poly(ADP-ribose)polymerase in critical disease

Abstract:Poly(ADP-ribose)polymerase(PARP)plays a vital role in DNA repair and genome stability in eukaryotic cells. Over-activation of PARP is involved in the pathophysiological processes of gene regulation, signal transduction and cell apoptosis, and plays an important role in the process of acute inflammation. This paper reviews the structure and biological function of PARP, and study prog-ress of PARP as a therapeutic target in several acute severe diseases such as ischemia-reperfusion inju-ry, multiple organ failure, shock, and acute pancreatitis. Besides, it also summarizes the roles and possi-ble mechanisms of PARP and its inhibitors in acute critical illness.
