Effects of penehyclidine hydrochloride on severe acute pancreatitis associated acute lung injury through TLR4 in rats

Abstract:Objective To investigate whether the effect of penehyclidine hydrochloride (PHCD)on acute lung injury induced by acute severe acute pancreatitis (SAP) is through TLR4 in rats.Methods Thirty-six healthy adult male SD rats were used and randomly divided into 3 groups,group A of sham operation,group B of pancreatitis-associated acute lung injury (PALI) and group C of PALI with PHCD.Rats of group B and group C were established the model of acute lung injury associated with SAP by retrograde injection of 4% sodium taurocholate into biliopancreatic duct.Rats of group C of acute lung injury with SAP were immediately given PHCD (0.2 mg/kg) after SAP.Rats of group A and group B were injected of the same amount of 0.9% sodium chloride solution.After modeling,the rats were sacrificed at 12 h.The wet/dry weight ratio of lung tissue was calculated.The expression of TLR4 in lung tissue was detected by Western Blot expression.Immunohistochemistry detection of lung tissue NF-κB p65 protein was observed under light microscope.Pathological changes of pancreatic and lung tissues were scored,the IL-1β,IL-6 and serum amylase were detected by ELISA.Results There were extensive infiltration of neutrophils,alveolar hemorrhage and necrosis and fat necrosis with pancreatic tissue of rats in group B and group C.These pancreatic tissue injury score were significantly higher than that of group A (P < 0.01).However,there was no statistically significant difference between the level of serum amylase in group B and group C (P > 0.05).The W/D raio of lung tissue in rats of group B and group C was significantly higher than those in group A (P < 0.05).Compared with the rats of group B,the lung tissue pathological changes of C group were significantly improved,and the lung W/D value was significantly lower than that of group B (P < 0.05).Compared with group A,the expression of TLR4 in lung tissue of group B and group C was significantly higher (P < 0.01),and the expression of TLR4 in group C was significantly lower than that in group B (P < 0.05).The lung levels of NF-κB p65,IL-1β and IL-6 expression in group B and C group were significantly higher than those in A group (P < 0.01).Moreover,the expression of NF-κB p65,IL-1β and IL-6 of group C was significantly lower than that in group B (P < 0.05).Conclusion Although the damage of pancreatic tissue of SAP was not alleviated by the PHCD,it might be possible to reduce the acute lung injury induced with SAP by TLR4 through NF-κB,IL-1β and IL-6 inflammatory factors.
