Effects of thromobopoietin on bone marrow stem cells and immune of thrombocytopenia mice induced by lipopolysaccharide

Abstract:Objective To investigate the effects of thromobopoietin(rhTPO) on bone marrow stem cells and immune of thrombocytopenia mice induced by lipopolysaccharide( LPS) . Methods 100 <br> male SPF C57/BL6 mice were randomly divided into five groups: healthy control ( C) group, model ( LPS) group ( intraperitoneal injection of LPS 30 mg/kg) , low, medium and high dose rhTPO ( L, M, H) treatment group ( continuous subcutaneous injection of different doses rhTPO for 3 days ) . The peripheral blood platelet count( BPC) and the mortality of 72 hours of every group were determined. The concentrations of tumor necrosis factor-α(TNF-α) and interleukins (IL-6, IL-10)in plasma on the fourth day were evaluated by ELISA. The CD34 + and CD41/CD61 of bone marrow cells were determined by flow cytometry. The organ indexes of thymus, spleen were calculated. Results Compared with C group, BPC of LPS,M and H group were obviously decreased(P<0. 05). BPC of L group was significantly higher than that of LPS, M and Hgroup on the third and fourth day (P<0. 01). M, H group had no increased than LPS group(P> 0. 05). The bone marrow megakaryocytes CD41/CD61 of LPS, L,M and H group on day 4were obviously elevated compared with that of C group( P<0. 01). The bone marrow megakaryocytes CD41/CD61 of L group was significantly higher than that of M, H, LPS group(P<0. 01). The bone marrow CD34+ expression of LPS, M, H group had no change compared with C group (P> 0. 05),but CD34+ expression in L group was increased than that of M, H, LPS group (P<0. 01). The plasma concentration of TNF -α,IL -6 on day 4 had no significant difference between groups (P>0. 05). The IL -10 concentration of L group was higher than that of other groups(P<0. 01). The thymus index of LPS, M, H group were lower than that in group C (P<0. 01),but it was increased in L group than that in LPS group(P<0. 01). 72 h mortality in L group was significantly lower than that in the other three groups (P <0. 01). Conclusion Low doses of rhTPO could improve the platelet count and reduce the 72 h mortality through promoting the proliferation of marrow megakaryocyte and stem cells,raising the level of anti-inflammatory cytokine and strengthening the immune when used in thrombocytopenia mice induced by LPS.
