Protective effect of Quercetin on hepatocytes in septic rats

Abstract：Objective To observe the protective effect of different dosage of Quercetin ( Que) on hepatocytes in septic rats, and provide evidence for clinical practice. Method s Septic rat model was established using SD rats by cecal ligation and puncture ( CLP) method.SD rats were randomly divided into 3 groups(n=8): sham group; CLP group; CLP+Que1 group; CLP+Que2 group.Rats in CLP group received intraperitoneal injections of 0.9%saline(10 mL/kg) after surgery and 8 h after surgery. In CLP+Que1 group, Que 50 mg/kg ( in 10 mL/kg 0.9% saline) was injected after surgery and 8 h after surgery.In CLP+Que2 group, the dosage of Que was 100 mg/kg.Blood samples were collected 12 h after surgery to examinethe levels of ALT, AST, PCT and TNF-α.Then the rats were sacrificed to collect intestine tissue samples.Pathological changes of intestine were observed under microscopy. Results In sham group, the hepatic lobule structure was complete and the shape was normal.In CLP group, hepatic cells swelling obviously, central venous and sinus hepaticus congestion, inflammatory cell infiltration, exhibiting slappy necrosis.Compared with the model group, indicators of liver tissue damage were improved significantly in CLP+Que groups, and especially in CLP+Que2 group.Compared with the sham group, levels of liver enzyme, PCT and TNF-αwere significantly elevated in other groups <br> (P<0.05), especially in CLP group.In two CLP+Que groups, levels of AST and TNF-αwas lower in Que 2 group (P<0.05), while no obvious differences of ALT and PCT existed between two groups (P>0.05).Conclusion Quercetin can improve the damage of hepatocyte in septic rats.It plays a protective role in hepatocyte function and high-dose group shows better results than low-dose group.
