A relationship study between anti -leukotriene treatment and leukotriene metabolism level, expression of cysLT1 receptor gene mRNA in patients with bronchial asthma
Abstract：Objective To analyse the correlation between the concentration of urine leukotrienes and cysLT1 receptor gene mRNA expression levels in the patients with bronchial asthma to establish the clinical index which could forecast the clinical effect of anti -leukotriene treatment by detecting the difference of urine leukotrienes between pre-therapy and post-treatment and the cysLT1 receptor gene mRNA expression levels in peripheral white blood cells.Methods 85 cases of patients with bronchial asthma were treated with monteukast sodium in five-week trial, usage of β2 -receptor agonist, pulmonary function, PEF and asthmatic symptom score were measured.The expression levels of cysLT1 receptor mRNA were analyzed semi-quantitatively by reverse transcription PCR ( RT-PCR ) , concentration of urinary leukotrienes between pre -therapy and post -treatment were detected by ELISA, then the relationship was compared between the changes of leukotriene metabolism level and its cysLT1 receptor gene mRNA expression level with different curative effects.Results A positive correlation was found between the concentration of urinary leukotriene before treatment and the cysLT1 receptor mRNA expression levels in patients of the effective group, both of the two index were significantly increased than ineffective group, the PEF level before treatment was contrary.Conclusion The concentration of urinary leukotriene and the cysLT1 receptor gene mRNA expression levels might be used as marks for anti-leukotriene treatment, and the concentration of urinary leukotriene has an important reference value in clinical response assessment.
