The protective effect of Ad-KGF against oxidative stress induced by hypoxia in A549 cells
Abstract:Objective To investigate the prevention of KGF gene adenovirus ( Ad-KGF) from oxidative stress injuries in hypoxic cultured A549 cells.Methods Ad-KGF-transfected A549 cells were treated with hypoxia, then divided in the prevented control group, prevention group, the treated control group and treat group.Every group was divided into 12 h, 24 h, 36 h phases.The KGF expression levels, LDH activity, the activity of NOS activity and ROS of cell culture supernatants were detected by ELISA.The mRNA expression levels p53 and Bax in A549 cells treated with hypoxia was detected by RT -PCR.Results The results showed that compared with the control groups, the prevention and treatment groups showed significantly increased the KGF concentrations in culture supernatant (P<0.01), significantly reduced ROS activity (P<0.01), significantly increased NOS activity (P<0.01), and significantly lowered LDH activity ( P<0.01); while the gene expression level of p53 and Bax in prevention and treatment groups was significantly lower than the control group. Conclusion The results suggest that KGF plays antioxidant and anti -apoptotic roles in hypoxic treatment of A549 cells.
