Effect of ERK signaling pathway inhibitor PD98059 on prolonged survival and reactive oxygen species in brain tissue of rats after cardiopulmonary resuscitation
Abstract:Objective To explore the effects of ERK signaling pathway inhibitor PD98059( PD) on prolonged survival and reactive oxygen species ( ROS) in brain tissue of rats after cardiac arrest/cardiopulmonary resuscitation ( CA/CPR) .Methods First of all, ventricular fibrillation ( VF) in rats were induced by transoesophageal ventricular pacing, establishing the model of CA/CPR.Thirty-two Sprague Dawley (SD) rats were randomly divided into two groups (n=16), administrated PD (0.3 mg/kg, treatment group) or saline (control group), respectively.The survival time was monitored in the following 72 hours after restoration of spontaneous circulation ( ROSC) .Secondly, Seventy-two SD rats were randomly divided into three groups ( n=24 ) , sham, control and PD group, each of which, after ROSC was arranged to detect ROS level in brain tissue with laser confocal microscope at 12, 24, 48 and 72 hour ( n=6) time point.VF in rats were induced except sham group, and medicated with former parts.Results ①The survival time in the treatment group was(44.25 ±23.28) hours, which was significantly longer than the control group [(25.31 ±23.52)hours, P<0.05].The cumulative survival rates in the treatment group were increasing, compared with control group after ROSC at 12, 24, 48 and 72 hour time points (P<0.05).②Treatment group and sham group had a lower ROS level at 12, 24, 48 and 72 hour time points compared with control group(P<0.01).ROS reached a peak at 24 hour in the treatment and control groups.Conclusion ERK signaling pathway inhibitor PD could prolong the survival time of rats after CPR, obviously decrease ROS level in brain, and promote the survival of animals after ROSC.
