Abstract:Objective To investigate the effect of hydrogen sulfide on PD -1 expression in the lung of rats with experimental pulmonary fibrosis .Methods Thirty six male SD rats were randomly divided into three groups:the control group , the pulmonary fibrosis group and the treatment group .The model of pulmonary fibrosis was produced by bleomycin endotracheally , while the control group was given saline under the same condition .From the second day the rats in the treatment group were treated with 3.12 mg/kg NaHS ( total volumn of 2 ml) ig, while the other two groups'rats were treated with 2 ml saline ig instead .On the seventh day and the twenty -eight day , the lungs were isolated .Pathological changes in the lungs were evaluated by HE stain and Masson's trichrome stain .The mRNA expression of PD-1 was evaluated by quantitative RT -PCR analysis , and the immunohistochemical method was used to investigate the change of PD -1 protein. Results The degree of alveolitis and pulmonary fibrosis in the treatment group was alleviated compared with that of the model group .The expression level of PD -1 and mRNA in the treatment group was significantly decreased compared with the model group ( P<0.05 ) .Conclusion Over-expression of PD-1 may be an important mechanism in the pathogenesis of pulmonary fibrosis .Hydrogen sulfide might play pivotal role in curing the experimental pulmonary fibrosis in SD rats by suppressing excessive expressing of lung PD -1.
