Changes of cyclooxygenase isoenzyme and effects of meioxicam in acute lung injury induced by lipopolysaccharide
Abstract:Objective Fo investigate the changes of cyclooxygenase(COX) isoenzym e and effects of meloxicam(MEL) in acute lung injury(ALI). Methods 96 Wistar rats were randomized into control, lipopolysaccharide(LPS) stimulated, MEL treated group, the changes of COX- 1 and COX- 2 protein were examined with immanohistochemistry, the levels of PGE2(served as COX activity indireactly) were measured by radio - immunoassay. Results No marked difference of COX - 1 protein was found in both LPS stimulated group and control group (P > 0.05), but COX- 2 protein expression and levels of PC, E2 in LPS stimulated group were rapidly increased and significantly higher than those in control group (P < 0.01 ). The peak value of COX- 2 mRNA and protein in ALI was at 4 h and 6 h, respectively. No significant changes in COX - 1 and COX - 2 protein expression were found between MEL group and LPS treated group ( P > 0.05), but the levels of PGE2 in MEL treated group were markedly lower than those in LPS treated group ( P < 0.01 ). Condusion Excessive expression of COX - 2and inappropriate activation of COX - 2 may play an important role in the pathogenesis of LPS - induced ALI, selective inhibition of COX - 2activity in MEL treated may down- regulate the inflammatory response in ALI.
