Study on the osteogenic properties of calcined bone materials loaded with tranexamic acid

Abstract:Objective To investigate the effect of tranexamic acid(TXA)in bone regeneration by in vitro and in vivo experiments.Methods The effects of different concentrations of tranexamic acid on the proliferative activity of MC3T3-E1 cells were detected by CCK-8 assay.The effects of different concentrations of tranexamic acid on the differentiation of MC3T3-E1 cells in vitro were detected by ALP quantitative assay;Calcined bone was prepared,loaded with tranexamic acid,and subjected to the experiment of repairing mandibular bone defects in rats,which was carried out by HE staining,Masson staining,and immunohistochemical staining of CD31,to evaluate the repair of bone defects and the angiogenesis of defect areas.Results Cell proliferation assay showed that on the first day,3.2 μg/ml,16.0 μg/ml and 80.0 μg/ml tranexamic acid significantly promoted the proliferation of MC3T3-E1 cells;10 000 μg/ml tranexamic acid significantly inhibited the proliferation of MC3T3-E1 cells.On day 2 and 3,10 000 μg/ml tranatemic acid significantly inhibited the proliferation of MC3T3-E1 cells;ALP quantitative assay showed that TXA had no significant effect on ALP activity of MC3T3-E1 cells.In animal experiments,HE and Masson staining showed that the TBC group had a small amount of bone repair only on the edge of the defect,and TBC-TXA group had formed osseous repair in most areas.Masson staining results showed that only a small amount of collagen was formed in the TBC group,while a large amount of collagen was formed in TBC-TXA group.CD31 immunohistochemical staining showed that only a small number of new blood vessels were distributed in the defect area of the TBC group,while many new blood vessels were formed in the defect area of TBC-TXA group.Conclusions Tranexamic acid promotes proliferation of MC3T3-E1 cells in vitro and repair of bone defects as well as angiogenesis in vivo.
