Analysis of GINS gene family as poor prognosis biomarkers for liposarcoma based on expression profile data

Abstract:Objective To explore the impact of GINS gene family on the prognosis of liposarcoma based on expression profile data.Methods GSE159659,GSE21122,and GSE30929 related with liposarcoma were downloaded from Gene Expression Omnibus(GEO)database.The differential expression of GINS gene family between liposarcoma and adipose tissues was analyzed using ggplot2 package of R language,and Kaplan-Meier survival analysis was performed utilizing the survival and survminer packages.Subsequently,Spearman's test was used to analyze the correlation between GINS gene family and multiple immune checkpoints in liposarcoma.The STRING database was used to screen the genes co-expressed with GINS1,GINS2,and GINS4.Finally,clusterProfiler package was used to perform functional enrichment analysis for the co-expressed genes.Results The expression level of GINS1 in liposarcoma tissue(5.11±1.01)was significantly higher than that in adipose tissue(4.05±0.25)(P<0.0001),the GINS2 expression(5.70±0.72)was significantly higher than that in adipose tissue(5.10±0.17)(P<0.001),and the GINS4 expression(5.33±0.48)was significantly higher than that in adipose tissue(4.86±0.33)(P<0.01).The distant recurrence free survival(DRFS)of liposarcoma patients with high GINS1(33.63±34.47 months)was significantly shorter than those with low GINS1(41.73±26.95 months)(P<0.0001).The DRFS of patients with high GINS2(35.44±34.07 months)was significantly shorter than those with low GINS2(39.91±27.87 months)(P=0.025),and the DRFS of patients with high GINS4(34.68±35.02 months)was significantly shorter compared to those with low GINS4(40.68±26.51 months)(P=0.0076).The immune checkpoint CD47 in liposarcoma was significantly positively correlated with GINS1(R=0.40,P<0.0001),GINS2(R=0.40,P<0.0001),and GINS4(R=0.33,P<0.0001).The immune checkpoint CTLA4 was significantly positively correlated with GINS4(R=0.30,P<0.001).Fifty genes significantly co-expressed with GINS1,GINS2,and GINS4 were screened through the STRING database,and these genes were involved in biological function of DNA replication and cell division,and related pathways.Conclusions GINS1,GINS2,and GINS4 could serve as poor prognostic biomarkers and effective therapeutic targets for human liposarcoma.
