Effects of terminal γ-ray irradiation sterilization on the viral inactivation and biomechanical property of the tendon allograft

Abstract:Objective To investigate the effects of terminal γ-ray irradiation sterilization on the viral inactivation and biomechanical property of the tendon allograft. Methods Cell culture was applieol to observe the inactivation effects of PRV, VSV, PPV, EMCV, HIV-1 on the tendon allograft by γ-ray irradiation ( 25 kGy ). Twelve hand tendons from healthy male adults were obtained. All tendons were randomized into 2 groups: the experimental group ( irradiation ); the control group ( fresh tendon, no irradiation ). Ultimate tensile load test was performed between the 2 groups. Results After irradiation processing, the virus titer declined at least 4 log and no virus was detected after three-generation passages. And there were no significant differences in the tensile load of the allogeneic tendons ( P > 0.05 ). Conclusions Terminal irradiated sterilization ( 25 kGy ) is an effective method for the virus inactivation of the allogeneic tendon, and the processing does not affect the tensile load and structure of the tendon.
