An exploratory study of second-line treatment of bone pain in the patients of metastatic breast cancer with a loading dose of ibandronate

Abstract:Objective To evaluate the effectiveness and safety of second-line treatment of bone pain in the patients of metastatic breast cancer with a loading dose of ibandronate.Methods A prospective observational study was conducted in 130 patients with advanced and metastatic breast cancer combined by moderate-severe bone pain who were admitted and received second-line treatment with a loading dose of ibandronate from January 2010 to April 2014. The pain score, consumption of analgesic drug, Karnofsky performance status ( KPS ) score were recorded before and after the treatment in all the patients. Meanwhile, their blood biochemical indices and safety were documented.Results In the ifnal analysis, 126 patients were included. When compared with the scores before the treatment, the pain scores were signiifcantly reduced (P<0.01 ) and the KPS scores were signiifcantly increased (P<0.01 ) at 1, 2 and 3 weeks after the treatment. And meanwhile, the administration dose of analgesic drug ( oxycodone ) was obviously reduced at 1, 2 and 3 weeks after the treatment when compared with the dose before the treatment (P<0.01 ). Bone pain was effectively alleviated in 89 patients, and the remission rate was 70.6%. The effective analgesic time in the patients with moderate pain was signiifcantly shorter than that in the patients with severe pain (P<0.05 ). Besides, few adverse drug reactions and no obvious liver or renal toxicity were observed among the 126 patients.Conclusions The second-line treatment of bone pain with a loading dose of ibandronate in the patients with metastatic breast cancer is effective and well tolerated. The patients’ quality of life will be improved and the amount of analgesic drugs will be reduced.
