Abstract: Objective To prove into the relative analysis of body mass index ( BMI ) and predisposing genes of knee osteoarthritis ( OA ) in Han population, so as to study the influences of BMI on the invasion of knee OA, thus conduce to understand the etiology of knee OA and the clinical prevention and treatment. Methods We chose 184 knee OA patients from our research center whose kellgren/lawrence ( K/L ) score were above II grade and 400 normal as age-matched control group, then measured the BMI. We used SPSS15.0 statistics software to analyze the distribution of BMI in control group and knee OA group, then calculated the percentage and odds ratio of obese persons, who carried C allele in the site of+104T/C in the 5’ non-translational region of GDF5 gene and 14 aspartic acids ( D14 ) in aspirin gene, in control group and knee OA group. Results Male BMI of control group and knee OA group were ( 22.4± 6.19 ) kg/m2, ( 25.2±3.27 ) kg/m2, female were ( 21.3±6.08 ) kg/m2, ( 25.0±4.12 ) kg/m2. The BMI difference of two groups had statistical significance ( P<0.01 ). However, no dignificant differences were found between male and female patients. The percentage of obese persons who carried C allele in the site of+104T/C in the 5’ non-translational region of GDF5 gene were respectively 33%, 42.3%in control group and knee OA group, and OR=0.51, P<0.01;those who carried D14 in aspirin gene were respectively 27.5%, 42.9%, and OR=1.98, P=0.01. Conclusions With the interaction of genetic factors, mechanical factor induced by obese educes important effect in the invasion of knee OA. Obese is a high-risk factor of knee OA.
