Isolation and molecular characteristics of hantavirus from hemorrhagic fever patient with renal syndrome in Heilongjiang province

Abstract:Objective To study the molecular characterization of a strain of hantavirus isolated from a patient with hemorrhagic fever with renal syndrome ( HFRS) in Heilongjiang province and its corresponding relationship with hantaviruses in rats. Methods A strain of hantavirus was isolated from the serum sample of a HFRS patient, and named WCL. Then the S gene of the virus was amplified, sequenced and analyzed. Results The complete nucleotide sequence of the S gene of WCL strain was composed of 1698 nucleotides with TA-rich. The S gene contained one ORF, starts at position 37 and ends at position 1326, encoding for the N protein of 429 amino acid residues, and in line with Hantaan ( HTN) -based coding. Sequence comparison showed that WCL was more homologous to HTN viruses than the other hantaviruses, most similar to Bao14 isolated from Apodemus agrarius in Heilongjiang Province, and the amino acid sequence of N protein of WCL has the conserved amino acid sites of HTN viruses. Phylogenetic analysis showed that WCL was clustered into the group of HTN type viruses and was more similar to Heilongjiang Bao14 strain and HTN261 strain than to others, and belonged H4 subtype. So the homology and phylogenetic analysis reflected a certain degree of geographical aggregation characteristic of hantavirus. Conclusion WCL belonged the H4 genetic subtype of HTN viruses with Bao14 and HTN261.
