Abstract:To address the limitations of traditional high-fructose syrup production, such as low catalytic efficiency of conventional glucose isomerases, this study focused on the heterologous expression and optimization of a novel glucose isomerase,Pd-xylA,derived from Pantoea dispersa isolated from kiwifruit roots. The Pd-xylA gene was cloned and expressed in Escherichia coli BL21(DE3),followed by systematic optimization of induction conditions including temperature and IPTG(isopropyl-β-D-thiogalactopyranoside)concentration.The results demonstrated that the optimal induction conditions were 28℃and 0.1 mmol/L IPTG,under which Pd-xylA achieved the highest soluble expression level. This study provided theoretical support for the development of enzyme resources with enhanced catalytic activity and industrial applicability, thereby advancing the biotechnological production of high-fructose syrup.
