Study on the Composition Differences of Quercetin Substances in Chestnut Peel before and after Digestion by UPLC-MS/MS

Abstract:Flavonoids are secondary metabolites of plants,which can inhibit pancreatic lipase and have potential function of regulating lipid metabolism.Quercetin belongs to flavonols in flavonoids.Therefore,it is of great significance to study the digestive mechanism of quercetin and its glycosylated derivatives in vivo to improve the research and development of functional products regulating lipid metabolism.This experiment used a widely targeted metabolomics technique of ultra high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS)to analyze the composition changes of quercetinsubstances in chestnut peel after simulated human digestion system.The results showed that there was a significant difference in the content of quercetinsubstances in chestnut skin samples before and after digestion(P<0.05).Differential metabolite analysis showed that the content of quercetin substances in chestnut skin decreased significantly before and after digestion,with quercetin-3-O-β-D-glucopyranoside decreased by 94.08%chestnut pellicle and 90.88%chestnut shell after simulated gastrointestinal digestion,respectively.In summary,the stability of quercetinsubstances in chestnut peel was poor during digestion.In the future,nanoparticle loading and other methods can be used to embed them to improve their bioavailability.
