Study on Genotype Characteristics of Measles Virus Epidemic Strains in Dingxi from 2011 to 2023

Abstract:OBJECTIVE To analyze the genotype characteristics of measles virus epidemic strains in Dingxi from 2011 to 2023.METHODS Pharyngeal swab specimens from suspected measles cases were collected and screened for measles virus nucleic acid positive by real-time RT-PCR.Viral RNA was extracted from positive isolates using Vero/SLAM cells.456 nucleotide fragments of measles virus N gene carboxyl terminal were amplified by RT-PCR,and sequenced and analyzed.RESULTS 1 134 throat swabs were collected from suspected measles cases in Dingxi from 2011 to 2023,182 nucleic acids positive,with a positive rate of 16.05%.39 measles virus strains were obtained by Vero-SLAM cell culture.The nucleotide sequences of 39 measles virus strains were analyzed and 39 measles virus strains belonged to H1a genetype.The genetic relationship showed that 39 measles virus strains belonged to the same branch as China93-2/H1a.39 wild measles strains were divided into two sub-branches,and both sub-branches had measles virus strains isolated from 2011 to 2018.The average genetic distance between 39 measles virus strains was 0.000~0.029.CONCLUSION The 39 measles virus strains isolated from Dingxi in Gansu from 2011 to 2023 are all H1a genotypes,which are consistent with the dominant epidemic strains in Gansu and the whole state.
