Exploring the Core Drugs and Molecular Mechanisms of Traditional Chinese Medicine for Treating Metabolic Related Fatty Liver Disease Based on Data Mining and Network Pharmacology Methods

Abstract:OBJECTIVE Based on data mining and network pharmacology methods,to explore the core drugs and molecular mechanisms of traditional Chinese medicine in the treatment of metabolic related fatty liver disease.METHODS Using the Tradition-al Chinese Medicine Inheritance Assistance Platform(V3.0)to construct a prescription database for traditional Chinese medicine treat-ment of MAFLD,mining core drugs,obtaining active ingredients and targets of traditional Chinese medicine using BATMAN-TCM,screening MAFLD related targets using OMIMand GeneCards,constructing an interaction network using Cytoscape 3.9.1,conducting PPI network analysis using STRING,further enriching analysis using DAVID,and docking with CB-Dock2 molecules,screening and validating the key targets and mechanisms of action of core drugs in the treatment of MAFLD.RESULTS 392 studies were in-cluded,involving 230 traditional Chinese medicine ingredients.The core drugs include:Poria cocos,hawthorn,Atractylodes macro-cephala,Zexie,etc.There are 317 effective active ingredients in the core drug,which can treat MAFLD through 341 potential tar-gets.860 biological functions and 189 signal transduction pathways were enriched,involving multiple processes such as longevity reg-ulation,insulin resistance,lipid and atherosclerosis.CONCLUSION Traditional Chinese medicine is commonly used with a combina-tion of cold and warm,sweet and bitter ingredients.It mainly regulates multiple pathways such as HIF-1,IL-17,AMPK,PPAR,FoxO through multiple targets such as AKT1,INS,ALB,IL6,TNF,TP53,PPARG,EGFR,IL1B,CTNNB1,etc.It intervenes in mul-tiple systems such as digestion,circulation,and endocrine,improves liver fat accumulation and immune inflammatory damage,and plays a therapeutic role.
