Methods for preventing non-specific punctate staining of CK(pan)in blank areas of glass slides and on tissue sections

Abstract:Objective: To investigate the impact of CK(pan) reacting with residual skin impurities on glass slides during immunohistochemistry and to explore solutions. Methods: Two brands of glass slides were used, and manual immunohistochemistry and MXTitan platform detection were performed in four groups: (1) Blank control group: handling blank glass slides with gloves. (2) Blank experimental group: handling 3 μm thick sections of tonsil tissue wax blocks with gloves and using glass slides for section retrieval. (3) Fingerprint control group: pressing fingerprints on designated areas of the glass slides. (4) Fingerprint experimental group: pressing fingerprints on designated areas of the glass slides and retrieving sections from 3 μm thick tonsil tissue wax blocks. Results: No spotty nonspecific staining was observed on the glass slides in the blank control group and blank experimental group. However, spotty nonspecific staining appeared on the glass slides with fingerprints in the fingerprint control group and fingerprint experimental group. Conclusion: During section retrieval, it is advisable to avoid touching the tissue retrieval area with fingerprints to prevent nonspecific staining from interfering with immunohistochemistry results.
