Application of a novel antigen retrieval solution in enhancing immunohistochemical staining effects on paraffin sections

Abstract：Objective To investigate the effect of a novel antigen retrieval solution on enhancing immunohistochemical staining of formalin fixed and paraffin-embedded tissue sections.Methods Three different antigen retrieval solutions(citrate retrieval solution,EDTA retrieval solution,and the novel retrieval solution)were applied to perform heat-induced high-pressure antigen retrieval on paraffin-embedded tissue sections from 30 different tissue samples.Subsequently,30 antibodies targeting antigens at different cellular localization sites(cell membrane,cytoplasm,and nucleus)were selected to perform immunohistochemical staining.The staining effects(including the intensity of positive staining and the number of positive cells)were observed,and statistical comparisons were made among the three groups.Results In the citrate retrieval solution group,staining intensity of+was observed in 30.0%(9/30)of samples,++in 66.6%(20/30),and+++in 3.3%(1/30).In the EDTA retrieval solution group,staining intensity of+was noted in 10.0%(3/30)of samples,++in 83.3%(25/30),and+++in 6.66%(2/30).In the novel retrieval solution group,staining intensity of+was found in 3.3%(1/30)of samples,++in 60.0%(18/30),and+++in 36.6%(11/30).Conclusion The novel antigen retrieval solution exhibits favorable retrieval effects in various tissue samples,it can shorten the antigen retrieval time,and provides a new option for tissue antigen retrieval,and has practical value.
