Relationship between tissues expression levels of lncRNA PCAT6 and lncRNA PCGEM1 and clinical pathological parameters as well as prognosis in lung adenocarcinoma

Abstract:Objective To investigate the relationship between the expression levels of long non-coding RNA prostate cancer associated transcript 6(lncRNA PCAT6)and long non-coding RNA prostate cancer gene expression marker 1(lncRNA PCGEM1)in lung adenocarcinoma tissues and clinicopathological parameters as well as prognosis of patients.Methods Tissue samples of lung adenocarcinoma and adjacent non-tumor tissues were collected from 100 patients with lung adenocarcinoma who underwent surgical resection at Yankuang New Mileage General Hospital between March 2016 and March 2018.Quantitative real-time polymerase chain reaction(qRT-PCR)was applied to detect the expression of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma and adjacent tissues,and the clinical data of patients were collected.The correlation between the expression of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma tissues was analyzed using Spearman correlation analysis.Kaplan-Meier method was applied to analyze the relationship between the expression of lncRNA PCAT6/lncRNA PCGEM1 in lung adenocarcinoma tissues and prognosis.The prognostic factors of lung adenocarcinoma patients were analyzed using Cox regression analysis.Results The expression levels of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma tissues were obviously higher than those in adjacent tissues(P<0.05).Using the mean expression levels of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma tissues as the cut-off values,patients were divided into two groups respectively:for lncRNA PCAT6,the high-expression group(55 cases)and the low-expression group(45 cases);for lncRNA PCGEM1,the high-expression group(52 cases)and the low-expression group(48 cases).The proportions of poorly differentiated tumors,lymph node metastasis,and TNM stage III in lncRNA PCAT6 and lncRNA PCGEM1 high-expression groups were obviously higher than that in the low-expression groups(P<0.05).Spearman analysis showed a positive correlation between the expression of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma tissues(r=0.539,P<0.05).Survival analysis results showed that the 5-year survival rate of patients with high expression of lncRNA PCAT6 and lncRNA PCGEM1 in lung adenocarcinoma tissues was lower than that of patients with low expression(P<0.05).TNM stage III,lymph node metastasis,poor tumor differentiation,and high expression of lncRNA PCAT6 and lncRNA PCGEM1 were independent risk factors for mortality in lung adenocarcinoma patients(P<0.05).Conclusion lncRNA PCAT6 and lncRNA PCGEM1 are highly expressed in lung adenocarcinoma tissues,and their high expression is also related to lymph node metastasis,TNM staging,and tumor differentiation.These two lncRNAs have good predictive value for the prognosis of lung adenocarcinoma patients.
