HER2 gene amplification status and its correlation with clinicopathological characteristics in 353 breast cancer patients with IHC 2+in Chengdu

Abstract:Objective To analyze the amplification status of human epidermal growth factor receptor 2(HER2)gene in breast cancer patients with immunohistochemistry(IHC)2+results in Chengdu,and to explore its correlation with clinicopathological features,so as to provide a reference for precise clinical diagnosis and treatment.Methods A retrospective analysis was conducted on 353 breast cancer patients with IHC 2+results who were admitted to Chengdu Fifth People's Hospital from January 2021 to January 2025.Fluorescence in situ hybridization(FISH)was used to detect HER2 gene amplification status,and statistical analyses were performed by combining with clinicopathological parameters,including patient age,maximum tumor diameter,histological grade,regional lymph node involvement,and hormone receptor status.Data on HER2 amplification rates in breast cancer from domestic and international studies were systematically reviewed,and statistical comparisons were made between these rates and the amplification rate obtained in this study.Results The positive rate of HER2 gene amplification detected by FISH was 35.13%(124/353),which was significantly different from that in other regions.Statistical analysis revealed that HER2 gene amplification was correlated with histological grade(χ2=4.8,rs=0.13,P<0.05),lymph node metastasis(χ2=32.03,V=0.304,P<0.001),and ER/PR-negative expression(χ2=5.10,V=0.117,P<0.05).However,there was no statistical between HER2 gene amplification and patient age or tumor size(P>0.05).Conclusion The HER2 gene amplification rate in breast cancer patients with IHC 2+is 35.13%,exhibiting regional variability and correlations with high histological grade,lymph node metastasis,and ER/PR-negative expression.
