Expression of m6A methyltransferase METTL3 and METTL14 and their significance in children with Wilms tumor

Abstract:Objective To explore the expression and significance of methyltransferase-like factor 3(METTL3)and methyl transferase-like factor 14(METTL14)in children with Wilms tumor.Methods A total of 94 children with Wilms tumor admitted to Handan Central Hospital were studied,and the cancer tissues and adjacent tissues at a distance of more than 2 cm from the lesion were collected intraoperatively,and the expression levels of METTL3 and METTL14 were detected in the tissues by fluorescence quantitative PCR and immunohistochemistry.The clinicopathological data of the children as well as the follow-up records were collected,and the relationship between the expression levels of METTL3 and METTL14 and the clinicopathological characteristics and prognosis of the children with nephroblastoma were analyzed.Results The expression level of METL3 in the cancer tissues of children with Wilms tumor was(1.60±0.28)and that of METL14 was(1.03±0.25),which were higher than that of METL3(0.92±0.16)and METL14(0.75±0.14)in the adjacent tissues(t=45.968,21.135,P<0.001);the high expression rates of METTL3(79.17%)and METTL14(84.38%)in cancer tissues were higher than those in adjacent tissues(34.38%)and 39.58%)(x2=39240,40.867,P<0.001).The expression levels of METTL3 and METTL14 were related to the clinical stage,lymph node metastasis and risk of the children(x2=5.250,6.829;4.796,5.349;12.924,10.818;P=0.022,0.009;0.029,0.021;0.002,0.004);High expression of METTL14,clinical stage Ⅲ to Ⅳ,lymph node metastasis,and high risk were risk factors that affected the prognosis(HR=1.407,1.325,1.370,1.264,P=0.025,0.039,0.027,0.002).The cumulative survival rate was lower in the METTL3 high-expression group(79.63%)than that in the METTL3 low-expression group(95.00%)(Log rank x2=4.465,P=0.035);the cumulative survival rate was higher in the METTL14 low-expression group(97.30%)than that in the high-expression group(78.95%)(Log rank x2=6.090,P=0.014).Conclusion METTL3 and METTL14 levels are significantly elevated in Wilms tumor in children cancer tissues,which are positively correlated,and both are associated with the prognosis of the affected children.
