Correlation of ACOT7 expression with molecular subtypes and clinicopathological features of breast cancer

Abstract:Objective To explore the expression characteristics of acyl-CoA thioesterase 7(acyl-CoA thioesterase 7,ACOT7)in breast cancer,to analyze its correlation with molecular subtypes and clinicopathological features of breast cancer,and to evaluate its potential as a prognostic biomarker.Methods Eighty paraffin-embedded tissues of invasive breast cancer(non-specific type)and 40 paraffin-embedded tissues of normal breast were collected from Wuzhou Red Cross Hospital,Guangxi.All patients did not receive anti-tumor treatment before surgery.The expression of ACOT7 was detected by using the immunohistochemical PowerVision two-step method.Patients were followed up by telephone or by collecting clinical data.The presence of light yellow to brown granules in the cytoplasm was considered positive,and the results were determined according to the proportion of positive cells and staining intensity.GEPIA was used to analyze differential gene expression,the Kaplan-Meier method was used to analyze survival prognosis,and SPSS 16.0 software was used for statistical analysis.Results The positive rate of ACOT7 in breast cancer tissues was 95%,which was significantly higher than that in normal breast tissues(27.5%,P<0.05),in which the with expression was localized in the cytoplasm.ACOT7 expression showed no significant correlation with patient age,tumor diameter,histological grade,TNM stage,ER,PR,and HER-2(P>0.05),but was significantly correlated with Ki-67(P<0.05).The positive rate of ACOT7 was the highest(100%)in HER-2 over-expression subtype and triple-negative breast cancer,followed by Luminal B subtype(95.56%).There was a significant difference in ACOT7 expression between the HER-2 over-expression subtype and the Luminal A subtype(P<0.05).ACOT7 was highly expressed in breast cancer tissues compared with normal breast tissues,and patients with high ACOT7 expression had a poorer prognosis(P<0.05).Conclusion ACOT7 is upregulated in breast cancer and correlated with Ki-67.Its high expression indicates a poorer prognosis,suggesting its potential as a prognostic biomarker.
